The play takes place mainly in Rome and its surrounding areas in 44 B.C.E., near the end of the Roman Republic.

The opening scenes are in the streets of Rome, where commoners celebrate Caesar’s triumphal return after defeating Pompey. Much of the central action stays in Rome: Brutus debates the conspiracy in his orchard, Caesar argues with Calpurnia at home, and the assassination takes place at the Capitol during a Senate gathering. Caesar’s funeral also takes place in Rome, where Brutus and Antony address the Roman crowd and Antony turns public feeling against the conspirators.

The later part of Julius Caesar moves away from Rome as civil war follows the assassination. Brutus and Cassius gather their forces in an army camp near Sardis, then march toward Philippi. The final act takes place on the battlefield near Philippi in Greece, where Brutus and Cassius's armies fight Antony and Octavius, leading to Cassius and Brutus's deaths.

This movement from Rome’s public streets and political institutions to distant battlefields follows the play’s pattern, as a political murder committed in the name of Rome spreads into civil war beyond the city itself.
